2. 所有动物细胞均视为有潜在的生物危害性,必须在二级生物安全台内操作,并请注意防护,所有废液及接触过此细胞的器皿需要灭菌后方能丢弃。

1)、细胞之接种密度为何?

 依照细胞株基本数据上之接种密度或稀释分盘之比例接种即可。细胞数太少或稀释的太多亦是造成细胞无法生长之一重要原因。

2)、悬浮性细胞应如何继代处理?

肺大动脉内皮细胞 一般仅需持续加入新鲜培养基于原培养角瓶中,稀释细胞浓度即可,若培养液太多时,可将培养角瓶口端稍微抬高,直到无法容纳为止。分瓶时取出一部份含细胞之培养液至另一新的培养角瓶,加入新鲜培养基稀释至适当浓度,重复前述步骤即可。

3)、冷冻管应如何解冻?

 取出冷冻管后,须立即放入37°C水槽中快速解冻,轻摇冷冻管使其在1分钟内全部融化,并注意水面不可超过冷冻管盖沿,否则易发生污染情形。另冷冻管由液氮桶中取出解冻时,必须注意安全,预防冷冻管之爆裂。

气道平滑肌是维持气道张力的根本。它存在于气管、支气管树直至终末支气管。其特异的超微结构特点和调节机制是实现气管正常生理功能的基础。有证据表明气管平滑肌在肺的发育过程以及诸如哮*病、慢性支气管炎和肺气肿等疾病状态下经历着显著的表型变化。另外,气道平滑肌通过分泌细胞因子和趋化因子等促炎介质,发挥免疫调节功能。培养的气管平滑肌细胞能表达三种P21活化的激酶(PAK)的同型异构体和PAK1,并通过P38 mapk信号途径参与气管平滑肌细胞的迁移。气道平滑肌细胞增殖已成为人们关注的焦点,因为在哮*病人中,此细胞是气道壁重塑反应中数量上的重要组分,并被认为是新型抗哮*病药物的作用目标。

    人气管平滑肌细胞 (HTSMC)提取于人气管组织,原代冻存。每管含有细胞数>5×105 cells/ml,此细胞通过对alpha-SMADesmin免疫荧光染色验证,经测试不含有HIV-1HBVHCV、支原体、细菌、酵母和真菌。细胞可以达到15倍增。

Description

 

Airway smooth muscle is an important tissue in maintaining airway tone. They exist in the trachea and in the bronchial tree up to the terminal bronchioles. Their specialized ultrastructural features and regulatory mechanisms are the basis of normal airway function. Evidence suggests that airway smooth muscle undergoes marked phenotypic modulation in lung development and in disease states such as asthma, chronic bronchitis and emphysema. Airway smooth muscle may also serve immunomodulatory functions, which are mediated by the secretion of pro-inflammatory mediators such as cytokines and chemokines [1]. Cultured tracheal smooth muscle cells express three p21-activated kinase (PAK) isoforms and PAK1 participates in tracheal smooth muscle cell migration by signaling to p38 Mapk [2]. Airway smooth muscle cell proliferation has been the focus of considerable attention, as it is a quantitatively important component of the airway wall remodeling response in asthma and has been suggested as a suitable target for the development of novel anti-asthma agents.

HTSMC from ScienCell Research Laboratories are isolated from human trachea. HTSMC are cryopreserved at secondary culture and delivered frozen. Each vial contains >5 x 10^5 cells in 1 ml volume. HTSMC are characterized by immunofluorescent method with antibodies to ?-smooth muscle actin and desmin. HTSMC are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HTSMC are guaranteed to further expand for 15 population doublings at the condition provided by ScienCell Research Laboratories.


Recommended Medium

 

It is recommended to use Smooth Muscle Cell Medium (SMCM, Cat. No. 1101) for the culturing of HTSMC in vitro.

HTSMC are for research use only. It is not approved for human or animal use, or for application in in vitro diagnostic procedures.


Storage

 

Directly and immediately transfer cells from dry ice to liquid nitrogen upon receiving and keep the cells in liquid nitrogen until cell culture needed for experiments.
