Description

BAFF is regularly detected by enzyme-linked immunosorbent assay in brain tissue lysates and in normal spinalFluid, and in astrocytes by doubleFluorescence microscopy. BAFF is localized in astrocytes close to BAFF-R-expressing immune cells. BAFF receptors are strongly expressed in situ in primary central nervous system (CNS) lymphomas. The TNF superfamily member B cell-activatingFactor (BAFF) plays an important role in humoral immunity and in autoimmune diseases, including RA. Local BAFF gene targeting inhibits proinflammatory cytokine expression, suppresses generation of plasma cells and Th17 cells, and markedly ameliorates joint pathology. The B cell activatingFactor BAFF (BlyS/TALL-1/zTNF4) is a tumor necrosisFactor (TNF)-related ligand that promotes B cell survival and binds to three receptors (BCMA, TACI, and the recently described BAFF-R). Human BAFF is mapped to chromosome 13q32-34. The standard used in this kit is recombinant soluble human BAFF (A134-L295) with a molecular mass of 19.6kDa.

ScienCells human BAFF ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. Human BAFF-specific polyclonal antibodies are pre-coated onto 8 x 12 divisible strips. The human-specific detection monoclonal antibodies are biotinylated. The test samples and biotinylated detection antibodies are subsequently added to the wells and then washed with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes to yellow after adding acidic stop solution. The intensity of yellow is proportional to the amount of human BAFF in the sample that is captured on the strips.
