It is a member of the ErbB proteinFamily, more commonly known as the epidermal growthFactor receptor (EGFR)Family. HER2/neu has also been designated as CD340 (cluster of differentiation 340) and p185. It is encoded by the ERBB2 gene. HER2 is a cell membrane surface-bound receptor tyrosine kinase and is normally involved in signal transduction pathways leading to cell growth and differentiation. It is encoded within the genome by HER2/neu, a known proto-oncogene. HER2 is thought to be an orphan receptor, as none of the EGFFamily of ligands able to activate it. However, ErbB receptors dimerize upon ligand binding, and HER2 is the preferential dimerization partner of other members of the ErbBFamily. The HER2 gene is a proto-oncogene located on the long arm of human chromosome 17 (17q21-q22). ScienCells Human ErbB-2 ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. Human ErbB-2-specific polyclonal antibodies are pre-coated onto 8 x 12 divisible strips. The human-specific detection polyclonal antibodies are biotinylated. The test samples and biotinylated detection antibodies are subsequently added to the wells and then washed with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes to yellow after adding acidic stop solution. The intensity of yellow is proportional to the amount of human ErbB-2 in the sample that is captured on the strips. Product Use This product is for research use only.
