*准确：能准确区分正常细胞、早期凋亡细胞、晚期凋亡细胞和坏死细胞，并计数各群细胞的比例。
 
使用说明 
 
1、 细胞收集与Annexin V-FITC结合
 
 （1）在完成细胞凋亡诱导处理后，1500rpm，5分钟离心后弃上清，收集细胞。（贴壁细胞收集：先用PBS洗涤贴壁细胞一次，再用胰酶消化液解离细胞。1500rpm离心5分钟，弃上清收集细胞。）
 （2）用PBS重新悬浮细胞，并计数。（如用含有EDTA胰酶细胞消化液消化细胞，建议将收集的细胞用PBS再洗涤一次。） 
 （3）取1～5×105细胞悬浮液，1500rpm，5分钟离心后弃上清，加入500ul的1×Binding Buffer。（用蒸馏水1:9稀释10×Binding Buffer）
 （4）加入5ul Annexin V-FITC，再加入10ul Propidium Iodide，轻轻混匀。
 （5）在室温下，避光反应5～15min。
 （6）在1小时内进行流式细胞仪或荧光显微镜检测。（如需更长时间保存，细胞可先用1×Binding Buffer洗涤后再用2%甲醛固定。固定的细胞混匀重悬即可进行检测。）
 
 
2、 流式细胞仪分析
 
 （1） 设定：激发波长Ex=488nm：发射波长Em=530nm。Annexin V-FITC的绿色荧光FITC通道为FL1；PI红色荧光通过PI通道为FL2。使用未经凋亡诱导的正常细胞作为对照，进行荧光补偿调节。
 
 
 
3、 荧光显微镜观察
 
 （1）如果用于荧光显微镜下检测，1500g离心5分钟，收集细胞。用50-100ul 1×BindingBuffer轻轻重悬细胞。取一滴细胞悬浮液于载玻片上，盖玻片观察。
 （2）采用双色滤光片，Annexin V-FITC荧光呈绿色，PI荧光呈红色。
 
注意事项 
 
1、 如果是贴壁细胞，也可直接用盖玻片培养和诱导细胞凋亡。
2、 Propidium Iodide （PI）有毒，需戴手套操作使用。
3、 本试剂盒适用范围为动物性细胞或组织。植物细胞组织，植物性海藻等不建议使用。
