MspI digests both nonmethylated and methylated DNA. HpaII is sensitive to CpG methylation. Methylated cytosines comprising CG dinucleotides within DNA remain unconverted following bisulfite treatment, whereas non-methylated cytosines are converted to uracil and detected as thymine following PCR.
