图1. 选择1 μg 或100 ng人胚肾上皮细胞HEK293总RNA、水稻幼苗总RNA、桃果实总RNA,用VAHTSTM mRNA-seq v2 Library Prep Kit for Illumina®分别构建插入片段大小为200-300 bp或150-200 bp的转录组文库。文库用Agilent 2100 Bioanalyzer进行分析。

表1. 1 μg 或100 ng universal human reference RNA分别用VAHTSTM mRNA-seq v2 Library Prep Kit for Illumina®以及Illumina TruSeq RNA Sample Preparation Kit v2构建文库。文库用Qubit定量后,按照相同浓度混样,用Hiseq 2500测序。可以看到,用Vazyme试剂盒构建的文库具有和Illumina非常近似的数据产出和数据质量。

图3. 1 μg 或100 ng universal human reference RNA分别用VAHTSTM mRNA-seq v2 Library Prep Kit for Illumina®以及Illumina TruSeq RNA Sample Preparation Kit v2构建文库,用Hiseq 2500测序,两两比较表达重复相关性,以spearman相关系数r表示。可以看出,1 μg (A)、100 ng (B)模板量下Vazyme VAHTSTM和Illumina TruSeq之间,以及用Vazyme VAHTSTM建库1 μg和100 ng之间 (C)的相关系数r均大于0.97,说明具有很高的表达重复相关性。

 

      VAHTSTM mRNA-seq v2 Library Prep Kit for Illumina®适用于起始模板为0.1-1 μg总RNA的Illumina平台转录组文库构建。本产品的模板适应性广泛, 对于植物样本具有很高的建库成功率。本产品不仅提供与Illumina TruSeq RNA Sample Preparation Kit v2相同的建库流程 (插入片段150-200 bp),还针对插入片段大于200 bp的文库提供优化的打断及分选条件。用本产品构建的文库在数据产出及数据质量上均与Illumina官方试剂盒保持高度的一致性。
