Tth DNA Polymerase

Tth DNA Polymerase(a)(Tth DNA聚合酶)是一种热稳定性酶，分子量为94kDa，从Thermus thermophilus HB-8中分离得到(1)。该酶在74℃时可进行DNA复制，在95℃的半衰期为20分钟。Tth DNA聚合酶在镁离子存在的条件下可催化核苷酸沿5`→3`方向发生聚合反应，形成双链DNA，也可在镁离子存在的条件下，以RNA为模板沿5`→3`方向发生核苷酸聚合反应。该酶也具有5`→3`外切酶活性。TthDNA聚合酶可用于PCR、RT-PCR、逆转录和较高温度条件下的引物延伸反应。10X Reverse Transcription Buffer: 100mM Tris-HCl (pH 8.3，25℃), 900mM KCl。10X Chelate Buffer: 100mM Tris-HCl (pH 8.3，25℃), 1M KCl,7.5mM EGTA, 0.5% Tween® 20, 50% 甘油。Thermophilic DNA Polymerase 10X Reaction Buffer: 500mM KCl, 100mM Tris-HCl (pH 9.0，25℃) 和1% Triton® X-100。缓冲液经过优化，适合与0.2mM的dNTP一起使用。氯化***和氯化镁: 提供10mM MnCl2和25mM MgCl2溶液。

储存条件: 储存于–20°C。

储存缓冲液: 10mM Tris-HCl (pH 7.5，25°C), 300mM KCl, 1mM DTT, 0.1mM EDTA, 0.5mg/ml BSA, 50% 甘油。

单位定义:在70℃条件下，30分钟内催化10nmol dNTP掺入到TCA不溶性物质所需的酶量为一个单位。
