dNTP用途

高质量的脱氧核糖核苷酸 (dNTP) 对 PCR 、 RT-PCR 、测序和标记等反应的成功都非常重要； PCR反应中 dUTP 替代 dTTP ，导致产物中含有尿嘧***，这种掺入适用于很多应用，将尿嘧***－ N －糖基化酶 UNG酶(有时称为UDG) 加入到 PCR 反应中，可切割任何污染PCR产物中的尿嘧***，从而避免错误的形成，这在临床 PCR 诊断试剂中应用非常广泛。如果四种脱氧核糖核苷酸的浓度不一致，将大大增加聚合酶的错配机率；如果纯度不高，或者混有甲基化的脱氧核糖核苷酸将造成产物的甲基化；或者含有脱氧核糖核苷一***酸、脱氧核糖核苷二***酸也会降低聚合酶的聚合能力。
