背景资料 AMCA 作为一种蓝色荧光标记，其吸收光波长最大为350nm，发荧光则为450nm。对于荧光显微镜来说，AMCA可以用*灯来激发，用紫外滤板来观察。由于 AMCA的信号相对较弱，单标实验中不推荐使用AMCA。AMCA和荧光素的荧光波长只有很小的重叠范围，而和发出长波长荧光的荧光基团没有或者只有极少 的重叠，因此它最常用于多标记实验中，比如免疫荧光显微镜和流式细胞仪。由于人眼不能很好的检测蓝色荧光，在多标记的实验中，AMCA耦联的二抗应当被用 于检测大量的抗原。AMCA和荧光素一样很快淬灭，使用抗淬灭剂可以减轻。且由于肉眼不能很好的检测AMCA所激发的蓝色荧光，因而AMCA耦联的二抗更 适用于检测大量存在的抗原。如果使用在流式细胞仪中，AMCA可以用*灯或者水冷却的氩光灯激发，因为它们发出的光线是在光谱的紫外区。

To detect Rat GM-CSF by sandwich ELISA (using 100μl/well antibody solution) a concentration of 0.5 - 2.0 μg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech’s Biotinylated Anti-Rat GM-CSF (500-P225Bt) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Rat GM-CSF.
Western Blot
To detect Rat GM-CSF by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 μg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant Rat GM-CSF is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
Neutralization
To yield one-half maximal inhibition [ND50] of the biological activity of Rat GM-CSF (0.5ng/ml), a concentration of 0.09–0.14 μg/ml of this antibody is required.
