GoTaq ® qPCR Master Mix (a,b,c) 是用于实时定量 PCR(qPCR) 的新型试剂系统。该试剂系统包含一种专利的 DNA 结合荧光染料，该染料与双链 DNA 结合后，能够产生强度高于 SYBR ® Green I 的荧光。该系统结合了以下诸多优势： GoTaq® 热启动酶、优化的缓冲液、专利荧光染料，可有效地提高实时定量 PCR 的可靠性、重复性和灵敏度。 GoTaq ® qPCR Master Mix 为即用型的 2X 预混母液，包括实时定量 PCR 反应所需的所有组分，样品 DNA 、引物以及稀释 DNA 标准品用的水除外。该混合母液经过处理，稳定性高，组要组分有：专利的双链 DNA 结合染料、低浓度的羧基 -X 罗丹明（ CRX ）参比染料（与常用的 ROX™ 染料功能、用法相同）、 GoTaq ® 热启动酶、 MgCl 2 、 dNTPs 、专利的反应缓冲液，可确保产生最佳的实时定量 PCR 结果。当用户使用某些类型的实时定量 PCR 仪时，需要较高浓度的参比染料，因此，该试剂盒还单独提供一管 100X CXR 参比染料，用于这些仪器的校准。

GoTaq ® qPCR Master Mix 的优势

在染色双链 DNA 时，本专利配方的染料比 SYBR ® Green I 产生更明亮的荧光。与 SYBR ® Green I 相比，该染料对 PCR 的抑制作用更小，使扩增更有效，在采用与 SYBR ® Green I 相同的波长检测条件和设置的情况下，通常能更早得到 C t 值，线性范围也更宽。 CRX 参比染料的检测波长和设置与 ROX™ 的相同。 GoTaq ® Hot Start Polymerase （ GoTaq ® 热启动聚合酶）为专利抗体封闭的全长的 Taq DNA 聚合酶，能够在室温下抑制酶的活性。将组装好的反应体系在 95°C 下孵育 2 分钟，即可激活该酶。专利的聚合酶 / 缓冲液配方能够支持较多的循环数目（ 45-50 个循环），也与那些需要较长激活时间（ 95°C 10 分钟）的热循环程序兼容。 GoTaq ® qPCR Master Mix 可用于任何一种能检测 SYBR ® Green I 或 FAM™ 染料的实时热循环仪。 GoTaq ® qPCR Master Mix 含有低浓度的 CXR 参比染料。如果您的热循环仪要求使用的参比染料浓度较高，请向反应体系中加入 100X 的 CXR 参比染料，使其终浓度为 1X 。
