Myelin oligodendrocyte glycoprotein(MOG)is a transmembrane protein, which is expressed exclusively in the CNS.MOG contains a single Ig-domain exposed to the extracellular space which allows autoantibodies easy access. MOG protein has been identified as a crucial autoantigen for multiple sclerosis in humans. MOG is capable to produce a demyelinating multiple sclerosis-like disease in experimental animals,namely experimental autoimmune encephalomyelitis (EAE) in rodents and monkeys. ·Description Recombinant Human MOG produced in E. coli is a single, non-glycosylated, polypeptide chain containing 121 amino acids and having a molecular mass of 14kDa, which includes its Ig-domain. ·Quality Control Biological activity: Female C57BL/6 mice (8-10 weeks old) were immunized (s.c.) with 50 μg/animal of human rMOG in complete Freund’s adjuvant followed by 300 ng/mouse of pertussis toxin on day 0 and day 2 (i.p.). Mice showed EAE symptoms. DA rats immunized with 50 μg/animal of human rMOG at the base of the tail (s.c.) displayed EAE symptoms as well. Purity: Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Amino-Acid Sequence:The sequence of the first five N-terminal amino acids was determined and was found to be Gly-Gln-phe-Arg-Val. Endotoxin:Less than 0.1ng/μg (1.0EU/μg) of rHuMOG as determined by LAL test.. ·Formulation Sterile and Lyophilized aliquots with a 0.2um filtered protein solution containing 20mM Hac-NaAc,pH4.0. ·Reconstitution It is recommended to reconstitute the lyophilized rHuMOG in sterile 18MΩ-cm H2O not less than 100μg/ml, which can then be further diluted to working solutions. ·Storage Lyophilized rHuMOG although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution recombinant human MOG should be stored at 4°C between 2-7 days and aliquots of reconstituted samples with 0.1% HSA or BSA are stable up to 3 months below -18°C for future use.
