C-C motif chemokine 4, also known as G-26 T-lymphocyte-secreted protein, Lymphocyte activation gene 1 protein, Macrophage inflammatory protein 1-beta, Small-inducible cytokine A4, T-cell activation protein 2, CCL4 and MIP-1B, is a secreted protein which belongs to the intercrine beta (chemokine CC) family. CCL4 is a chemoattractant for natural killer cells, monocytes and a variety of other immune cells. CCL4 is a major HIV-suppressive factor produced by CD8+ T cells. Recombinant CCL4 induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form CCL4 (3-69) retains the abilities to induce down-modulation of surface expression of the chemokine receptor CCR5 and to inhibit the CCR5-mediated entry of HIV-1 in T-cells. CCL4 (3-69) is also a ligand for CCR1 and CCR2 isoform B. ·Description Novoprotein Recombinant Human CCL4/MIP-1 beta produced by mammalian cells expression system.Target protein is expressed with sequence (Ala24-Asn92) of Human ccl4 (Uniprot #P13236) fused with a polyhistidine tag at the C-terminus. ·Quality Control Purity: Greater than 90.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Endotoxin:Less than 0.1ng/μg (1EU/μg) of CCL4/MIP-1 beta (Human) ·Formulation Novoprotein Recombinant Human CCL4 was lyophilized at 1mg/ml in 20mM PBS. ·Reconstitution We recommend a quick spin followed by reconstitution of Novoprotein Recombinant Human CCL4 in sterile 18MΩ-cm H2O not less than 100μg/ml, which can then be further diluted to working solutions. ·Storage Lyophilized Recombinant Human CCL4 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution Recombinant Human CCL4 should be stored at 4°C between 2-7 days and aliquots of reconstituted samples are stable up to 3 months below -18°C for future use. Please avoid freeze-thaw cycles. |
