Robust, High-Fidelity PCR Amplification PfuTurbo® DNA polymerase*,‡ is a special formulation of cloned Pfu DNA polymerase** and the exclusive ArchaeMaxx® polymerase enhancing factor*** that has been shown to significantly increase PCR product yields without affecting replication fidelity. PfuTurbo DNA polymerase exhibits an average error rate estimated to be 2- to 60-fold lower than other proofreading enzymes and 6- to 100-fold lower than Taq DNA polymerase, which lacks proofreading activity.1 PfuTurbo DNA polymerase can also amplify higher yields of complex targets than Taq DNA polymerase or any other commercially available proofreading enzyme. The enhanced performance of PfuTurbo DNA polymerase allows the use of shorter extension times, fewer PCR cycles and lower concentrations of DNA template than are required for Pfu DNA polymerase. PfuTurbo DNA polymerase also amplifies cDNA with high accuracy§. Exclusive ArchaeMaxx® Factor Stratagene scientists isolated the ArchaeMaxx factor from Pyrococcus furiosus and added it to our high-fidelity enzymes, where it eliminates a PCR inhibitor and promotes shorter extension times, higher yield and greater target length. The ArchaeMaxx factor is a key ingredient in many of Stratagenes enzyme formulations. The ArchaeMaxx factor improves the yield of products amplified with Pfu DNA polymerase by overcoming "dUTP poisoning", which is caused by dUTP accumulation during PCR through dCTP deamination1. This dUTP poisoning inhibits Pfu and many other archael proofreading polymerases, such as VentR® and Deep VentR® DNA polymerases, limiting their efficiency1,2. The ArchaeMaxx factor functions as a dUTPase, converting poisonous dUTP to harmless dUMP and inorganic pyrophosphate resulting in greatly enhanced overall PCR performance. PfuTurbo® Hotstart DNA Polymerase PfuTurbo® Hotstart DNA polymerase improves specificity, reduces background and enhances yield of challenging PCR systems. This hot start enzyme is ideal when detecting low-copy-number targets in complex DNA backgrounds. In addition, PfuTurbo Hotstart DNA polymerase provides improved specificity when amplifying systems prone to primer-dimer formation or when performing robotic PCR applications that require prolonged incubations at room temperature prior to temperature cycling. PfuTurbo Hotstart DNA polymerase is an antibody-inactivated DNA polymerase that can be introduced into PCR protocols with no modification to cycling parameters. In this unique hot start formulation, both the DNA polymerase and Higher Amplification Efficiency Amplification efficiency is the measurement of fold amplification per PCR cycle, and is expressed as a fraction or percentage relative to perfect doubling (a PCR resulting in perfect doubling would exhibit 100 amplification efficiency). Once the efficiency of a PCR system is determined, it can be used to measure fold amplification from a particular template amount, or alternatively, to calculate the number of PCR cycles needed to obtain a desired yield. A small change in amplification efficiency is magnified over a multiple-cycle PCR, causing significant changes in product yield. For example, in a 25-cycle PCR, a 5 improvement in amplification efficiency can translate to a two-fold increase in product yield.† Lower-efficiency reactions not only produce lower yields, but take longer to reach saturation than high-efficiency reactions, increasing the chances of generating primer-dimers and undesired side products. Using real-time QPCR methods, it was determined that PfuTurbo DNA polymerase exhibits higher amplification efficiencies than other proofreading polymerases3.
