Capto MMC Media/HiTrap Capto MMCTechnical Information
Capto MMC is a salt-tolerant multimodal cation exchanger for process-scale applications. Capto MMC gives increased productivity and reduced cost.
Capto™ MMC is a multimodal cation exchanger. The multimodal functionality gives a different selectivity compared with traditional ion exchangers. This includes binding of proteins regardless of ionic strength of the loading material (see Figure below). This means that the medium may be used for direct load of clarified feedstocks, without prior dilution to reduce the conductivity of the starting material. The novel selectivity of Capto MMC can also be used to solve specific purification problems, at high as well as at low ionic strength. HiTrap™ Capto MMC are prepacked 1-ml and 5-ml columns for screening of selectivity, binding and elution conditions, and method scouting. To reduce tedious sample preparation, the top filter in HiTrap Capto MMC has been optimized for loading sonicated unclarified cell lysate directly on the column. Dynamic binding capacity of Capto MMC at 1 min residence time for three different proteins at different conductivities shown as NaCl concentrationin loading buffer.
The Capto™ MMC ligand may interact with proteins in many different ways. It contains a carboxyl group, and thus it is a weak cation exchanger. However, in addition to the ionic interactions several other types of interactions are involved, including hydrogen bonding and hydrophobic interaction.
