Introduction to the Blood-Brain Barrier

TThe blood-brain barrier (BBB) explains why certain substances circulating in the blood are able to get into the brain and others do not. The BBB is formed by brain capillaries that look like simple tubes of endothelial cells. Experimental and clinical evidence suggests that the BBB maintains the chemical environment for neuronal function and protects the brain from harmful substances. The blood-brain barrier is therefore crucial for complex issues such as drug delivery, pathogenesis of chronic neurological diseases, and issues related to bio-defense. We have developed a practical in vitro model that accurately reflects BBB characteristics.

In contrast to other available models of the blood-brain barrier, Flocel's Dynamic In vitro Blood-Brain Barrier (DIV-BBB) respects the anatomical aspects of the in situ endothelial cell-astrocyte interactions and replicates the physiological levels of shear stress to which in situ endothelial cells are exposed. The DIV-BBB allows formation of physiological transendothelial resistance, and formation of gap junctions that can be easily visualized at the electron microscope or determined experimentally with the use of tracers.

In vitro BBB Technology

The blood-brain barrier (BBB) explains why certain substances circulating in the blood are able to get into the brain and others do not. As determined by electron microscopy, the BBB is formed by brain capillaries that look like simple tubes of endothelial cells linked by tight junctions. The Flocel in vitro BBB is a tool that allows to reproduce in a flow-based 3D cell culture apparatus the essential properties of the human BBB, including the expression of metabolic and extrusion mechanisms responsible for multiple drug resistance to antiepileptic drugs, anti-tumor agents, etc.

The Flocel apparatus consists of hollow fibers (the artificial blood vessels) that are surrounded by an extracapillary space (the brain side). In addition to the cerebrovascular system, this apparatus can also be used to mimic the bone marrow, the placental barrier, or the CSF-blood barrier. In the case of modeling of bone marrow, the abluminal space is an ideal environment for the growth and differentiation of stem cells.

The Flocel DIV-BBB package contains: in vitro cell culture cartridges, a computer-controlled perfusion system, and a trans-endothelial electrical resistance (TEER) measurement module.

In contrast to other available models of the blood-brain barrier, the DIV-BBB respects the anatomical aspects of the in situ endothelial cell-astrocyte interactions and allows the formation of tight junctions that can be easily visualized at the electron microscope or determined experimentally with the use of tracers. Cell differentiation is also crucially modulated by exposure to shear stress.

Trans-endothelial Electrical Resistance (TEER) Measurement Module


The Trans-Endothelial Electrical Resistance (TEER) measurement provides a quick and easy evaluation of the integrity of the Blood-Brain Barrier model. The DIV-BBB model has a TEER closer to that found in vivo, >1000 Ω-cm2as opposed to the monolayer model that typically achieves a TEER of 200 Ω-cm2.

The TEER Measurement System is sold with the measurement software, the required cable, and a mounting platform that can accommodate four DIV-BBB cartridges. The electrodes in the cartridge plug into the mating connectors in the platform. The system is shipped with four cartridges for initial evaluation.

Computer-controlled Perfusion System

The Flocel Quad Pump mimics the pulsitile action of the heart in this model. The Quad Pump contains four small, easily configurable, pulsitile pumps that allow continuous flow through the DIV-BBB model. Each pump is individually controlled, allowing up to four independent experiments to be conducted at one time.
