Product overview
The Pyruvate Assay Kit provides a simple, direct and automation-ready procedure for measuring pyruvate concentration in various biological samples such as blood, cells, culture and fermentation media, etc. In the assay, pyruvate is oxidized by pyruvate oxidase via enzyme reactions to generate colour and fluorescence. The colour or fluorescence intensity is proportional to pyruvate content. Thus the pyruvate concentrations can be accurately measured by either colorimetric (spectrophotometry at OD = 570 nm) or fluorometric (Ex/Em = 535/587 nm) methods. The kit detects 1-1000 µM pyruvate samples.
Notes
Pyruvate quickly used up in different metabolic reactions so we recommend using samples as soon as they are harvested or extracted. We also recommend to deproteinate samples to remove interfering enzymes using either PCA or our 10 kDa spin cut-off filters (ab93349) after which they can be aliquoted in working volumes and stored at -80C until further use.
