Calibrating bulk recombinant proteinsMany suppliers offer recombinant cytokines and chemokines for use in cell culture and other applications. In most cases, these reagents are supplied in quantities of 5 to 100µg per vial and sometimes include a packaging overfill. (A 20µg vial may actually contain 22µg.) Package claim is based on mass, not on immunoreactivity as measured in ELISA. In order to utilize a bulk recombinant for ELISA, it must be "value assigned". An assay is run using the Calibrated ELISA Standard to make the standard curve. The bulk recombinant protein is then diluted in the range of the standard curve and each dilution is read as an "unknown." The calculated concentration vs. the expected concentration will determine the value assignment of the starting bulk recombinant material. Example procedure for value assignment of bulk recombinant proteinsIf the starting material (bulk recombinant) is a 5µg vial and it is reconstituted with 500µL of diluent, then diluted 1:10 in a buffer containing carrier protein, the concentration is assumed to be 10µg/mL. If the assay range for the ELISA in development is 15 to 1,000pg/mL, the bulk recombinant would be diluted to 2,000pg/mL, 1,000pg/mL, 500pg/mL, 250pg/mL, 125pg/mL, and so on. Each dilution is treated as an unknown sample and evaluated in the ELISA based on the Calibrated ELISA Standard curve. The calculated concentration in ELISA vs. the expected concentration based on the label claim of the bulk recombinant will determine the value assignment. For example, the assumed dilution of 250pg/mL may actually read 400pg/mL in the ELISA (40% higher). In this case, the starting material assumed to be 250ng/mL would be "value assigned" at 400ng/mL. The dilution that falls on the most linear portion of the curve should be used to determine the value assignment of the bulk recombinant material. Value-assigned recombinant should be frozen in aliquots to avoid repeated freeze-thaw cycles. We offer a full line of Calibrated ELISA Standards for Inflammatory and Th1/Th2 cytokines.
