Product Overview

Stemgent® Sox2-11R is a full-length recombinant human protein fused to an eleven arginine (i.e., 11R) membrane penetration domain at the C terminus to enable penetration across the plasma membrane of mammalian cells. The protein was expressed in E. coli as inclusion bodies, solubilized, refolded, and further purified. The protein identity was confirmed by western blot analysis. The in vitro function was tested using specific DNA binding assays. 11R proteins were reported to successfully generate induced pluripotent stem (iPS) cells from OG2 MEFs1.

Usage NoteHongyan Zhou and colleagues in the Sheng Ding laboratory at The Scripps Research Institute demonstrated the use of recombinant transcription factor proteins to generate protein-induced pluripotent stem (piPS) cells1. Reprogramming with recombinant proteins is still an undeveloped technology and the subject of ongoing research investigations. Scientists in Sheng Ding’s laboratory and at Stemgent have developed an optimized suggested method based on the original publication. Due to the low efficiency, further optimization is required in the experimental design.
