Due to the mild deprotection conditions we recommend using fast deprotecting amidites. Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in sealed vial to ensure that all contents are dissolved. We recommend using Ethyl thiol tetrazole for the activator reagent with this product.

Cleave and deprotect the CAL dye labeled oligo, in the presence of the CPG support, in 2-Methoxyethylamine (Ald# 241067-50) and MeOH (1:3) 3 hours at 60 °C. Filter sample and evaporate supernatant. Re-dissolve the CAL dye labeled oligo in water. The use of strong bases, such as concentrated ammonia, causes degradation of the dye and should be avoided.

CAL Fluor® Red 590 dye is an amidite, which fluoresces in the yellow-orange region of the visible spectrum. CAL Fluor Red 590 amidite is used for the 5' labeling of fluorogenic probes used in 5' nuclease assays, Molecular Beacons, Scorpion® primers and other genomic assays. This amidite does not contain a DMT protecting group and can only be added to the 5' terminus of the oligo. CAL Fluor Red 590 fluorophore is an alternative dye for TAMRA and is quenched by BHQ®-2 dye.
