Treatment of trypsin with phenylmethylsulfonyl fluoride (PMSF) followed by a strong base leads to the conversion of Serine 195 to dehydroalanine at the active site.  Alpha and beta forms of anhydrotrypsin are separated by immobilized soybean trypsin inhibitor.
