Application Notes	Polyclonal type IV is useful for immunoblotting, immunocytochemistry on fixed cells (methanol fixation) and immunohistochemistry on frozen tissues (fixed in methanol, aceton or 1% paraformaldehyde) when using a PBS buffer containing 0.1 mM CaCl2 and 0.1 mM MgCl2. For immunohistochemistry on paraffin tissue the sections should be pretreated with 1 mg/ml pepsin in 0.01 N HCl for 30 minutes at room tempeRature. Optimal antibody dilution should be determined by titRation; 1:25-1:100 for immunohistochemistry with avidin-biotinylated Horseradish peroxidase complex (ABC) as detection reagent, and 1:100-1:1000 for immunoblotting appliCations.
