Background: The Myc signaling pathway plays an important role in cell proliferation, differentiation,
transformation and apoptosis. The c-Myc protein is a transcription factor that heterodimerizes
with Max to regulate transcription of genes involved in proliferation, transformation and
angiogenesis. Myc mutations have been linked to the development of a number of human
cancers, including Burkitt’s lymphoma, cervical, ovarian, breast, lung and pancreatic carcinoma,
making Myc a promising therapeutic target.
Description: The Myc Reporter – HCT116 cell line contains the firefly luciferase gene under the control of Myc responsive elements stably integrated into HCT116 cells, a human colon cancer cell line. HCT116 contains a mutated beta-catenin which leads to the accumulation of β-catenin and constitutive activation of downstream Myc that induces the expression of Myc luciferase reporter. The cell line is validated for the inhibition of the expression of Myc luciferase reporter.
Materials Required But Not Provided: ICG-001 (Selleckchem #S2662): an inhibitor of the Wnt/β-catenin pathway. Myc can be activated by the Wnt pathway. Supplied As: Each vial contains 1.5 X 106 cells in 1 ml of 10% DMSO.
Host Cell line: HCT116
Mycoplasma Testing: The cell line has been screened using the PCR-based VenorGeM® Mycoplasma Detection kit
(Sigma-Aldrich) to confirm the absence of Mycoplasma species.
Growth Media: Thaw Medium 7 (BPS Bioscience #60185): McCoy’s 5A medium (Hyclone #SH30200.01) with 10% FBS (Life technologies #26140-079), 1% Penicillin/Streptomycin (Hyclone SV30010.01). Growth Medium 7B (BPS Bioscience #79545): Thaw Medium 7 (BPS Cat. #60185) plus 400 µg/ml of Geneticin (Life Technologies #11811031). Cells should be grown at 37°C with 5% CO2 using Growth Medium 7B. Storage / Stability: Immediately upon receipt, store in liquid nitrogen.
Application(s): Monitor Myc pathway activity and ccreen for activators or inhibitors of the Myc pathway.
