50 µl reaction mix (50 mM Tris pH 8.8, 5 mM MgCl₂, 4 mM DTT, 0.05% Tween-20, 800 µM S-adenosyl methionine, and SUV4-20H1) is added to the wells coated with the substrate. Incubate for 4 hr. Add primary antibody 18, incubate 1 hr. Then, add secondary HRP-labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminescence.
