50 µl reaction mix (50 mM Tris pH 8.8, 5 mM MgCl₂, 4 mM DTT, 20 µM S-adenosylmethionine, and 0-40 ng SUV39H2) is added to the wells of a substrate-coated plate. Incubate for 1 hr. Add antibody against methylated K9 residue of histone H3, incubate 1 hr.Finally, incubate 1 hr. with the Europium-labeled secondary antibody. Time resolved fluorescence measured at room temperature (?exc=330 nm, ?em=620 nm).
