Background: PLK3 or Polo-like kinase 3 is a cytokine-inducible kinase that is a member of the Polo family of serine/threonine protein kinases. PLK3 contains both a catalytic domain and a putative regulatory domain and plays an important role in the regulation of cell cycle progression and tumorigenesis. PLK3 is closely associated with centrosomes in several human cell lines and its localization is dependent on the integrity of microtubules. Hence, PLK3 is also involved in regulating microtubule dynamics and centrosomal function.

Specific Activity: ≥110 pmol/min/μg
Assay Conditions: 40 mM Tris-HCl pH 7.4, 20 mM MgCl2, 0.1 mg/mL BSA and 1 mM DTT using 1 mg/ml Casein substrate and 20 μM ATP. Reaction was done at 30°C for 45 min. Amount of ATP transferred was calculated using Kinase-Glo reagent from Promega.
Formulation: 40 mM Tris-HCl pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, 3 mM DTT and 20% glycerol.
Storage / Stability: At least 6 months at -80°C. Application(s): Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
