Background: JNK2 is a member of the JNK family that acts as an integration point for multiple biochemical signals and is involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. JNK2 targets specific transcription factors, and mediates immediate-early gene expression in response to various cell stimuli. Both UV radiation and the proinflammatory cytokine TNFα can induce JNK2. JNK2 play a main role in the regulation of regional specific apoptosis (1) during early brain development and providing protection against autoimmune diabetes (2).
Description: Human JNK2 (GenBank Accession No. NM_139069), full length, with N-terminal GST tag, MW=~70 kDa, expressed in Sf9 insect cells via a baculovirus expression system.

Assay Conditions: 5 mM MOPS, 2.5 mM β-glycerophosphate, 5 mM MgCl2, 1 mM EGTA, 0.4 mM EDTA, 0.05 mM DTT, 2 mM ATP. Incubate JNK2 with ATF2
substrate and 50 ?M [33P]-ATP at 30°C for 15 minutes, then spot reaction on phosphocellulose paper, fix in 1% phosphoric acid, and assay with a scintillation counter.
Formulation: 50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 10 mM glutathione, 0.1 mM EDTA, 0.25 mM DTT, 0.1 mM PMSF, 25% glycerol.
