10 μl of varying amounts of PLK4 was mixed with 5 μl of 1 mg/ml MBP protein substrate and 5 μl of distilled water. Reaction was initiated by adding 5 μl of 250 μM [33P]-ATP and incubating for 15 min at 30°C followed by spotting the reaction on phosphocellulose paper, fixing in 1% phosphoric acid, and assaying with a scintillation counter.

50 mM Tris-HCl, pH 7.5, 150 mM NaCl, 10 mM glutatione, 0.1 mM EDTA, 0.25 mM DTT, 0.1 mM PMSF, 25% glycerol.

Aqueous buffer solution

>6 months at –80°C.

Useful for studying enzyme kinetics and screening small molecule inhibitors.
