Aqueous buffer solution.

BRD4 protein is incubated with biotinylated acetyl-histone peptide substrate 2.5 μM stock (BPS Bioscience #33000) in BRD assay buffer (BPS Bioscience #33001) in a 10 μl reaction for 1 hour at room temperature. AlphaLISA® Nickle acceptor beads (PerkinElmer) are added, followed by Alphascreen® Streptavidin-conjugated donor beads (PerkinElmer). After 1 hour of incubation at room temperature, Alphacounts are measured.

50 mM Tris-HCl, pH 8.0, 138 mM NaCl, 2.7 mM KCl, 0.05% Tween- 20, and 10% glycerol.

At least 6 months at –80°C.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
