BET Ligand (Cat. #33000) was incubated with un-tagged BRD4 for 15 min at room temperature in 1X BRD assay buffer. GST-tagged BRD4 (Cat. #31044) was added and incubated for 20 minutes followed by addition of 20 ?l mixture of GSH Acceptor beads and Streptavidin-Donor beads for AlphaScreen^® analysis. Total reaction mixture was incubated for 35 minutes and the AlphaScreen^® signal was measured by EnSpire^® Multilabel Plate Reader (PerkinElmer).

40 mM Tris-HCl, pH 7.5, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, and 20% glycerol.

Aqueous buffer solution

Store at -80°C. Stable for >6 months when stored as directed.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
