BRD4 is incubated with biotinylated acetyl-histone peptide substrate (BPS# 33000) in BRD assay buffer (BPS# 33001) in a 10 µl reaction for 1 hour at RT. AlphaLISA^® ® Nickel Chelate Acceptor Beads (PerkinElmer) are added, followed by AlphaScreen® Streptavidin-conjugated donor beads (PerkinElmer). After 1 hour incubation at RT, Alphacounts are measured.

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 1.28 µM imidazole, and 20% glycerol

Aqueous buffer solution

>6 months at –80°C.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
