MTAP activity was measured using a xanthine oxidase-coupled method. MTA cleavage by MTAP produce adenine, which is then converted to 2,8- dihydroxyadenine by xanthine oxidase, leading to an increase in absorbance at 305 nm. 50 µl reaction mixtures contained MTAP, 50 mM KHsub>2PO₄, pH 7.4, 200 µM MTA, 0.02 units Xanthine Oxidase, and 1 mM DTT. Activity was measured at 37°C by continuous monitoring the absorbance at 305 nm for 30 min.
