Assay Conditions: Assay Conditions: All of the reactions were conducted at room temperature. The 100 µl reaction mixture in TDO2 Assay Buffer contains 50 nM TDO2, 500 μM tryptophan, and the coupled reaction components. The reaction mixture incubated for up to 120 min while reading the UV absorption signal. Absorption signals were measured using a Tecan Infinite M1000 plate reader. Final Assay Conditions: 0.1M Phosphate buffer, pH 7.0 0.01% Tween-20 0.1 mM Ascorbic Acid 0.16 µM Hematin 0.56 mM L-Tryptophan Formulation: 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween 20, 20% glycerol, 250 mM Imidazole
