Reaction was performed in 50 mM Tris pH 7.4, 5 mM MgCl2, 1 mM DTT, 5 µM ATP, 10 µM Ub, and 0.05% Tween. Reaction was initiated with the addition of UBE1 and was incubated at 30°C for 45 min. The luminescence was read with the Kinase Glo Max reagent (Promega).

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, 100 µg/mL FLAG peptide, 3 mM DTT and 20% glycerol.

Aqueous buffer solution

>6 months at -80°C.

Useful in conjunction with E2 and E3 for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
