Background: ALK1 is a serine/threonine-protein kinase receptor R3 precursor that mediates signal by TGFβ superfamily (1). ALK1 functions as a TGFβ type I receptor in endothelial cells and is responsible for human hereditary hemorrhagic telangiectasia (HHT) type II. Distinct Smad proteins (i.e., Smad2/Smad3 and Smad1/Smad5) show interaction with ALK1 and mediate TGFβ signaling. Northern blot and RT-PCR analysis show that ALK1 specifically induces expression of Smad6, Smad7, Id1, Id2, endoglin, STAT1, and interleukin 1 receptor in endothelial cells. ALK1 expression in inflammatory myofibroblastic tumor of the urinary bladder have also has been reported (2).
Description: Recombinant human activin receptor-like kinase 1, or ALK1, (a.a. 144-end), with N-terminal GST-tag, expressed in Sf9 insect cells via a baculovirus expression system. The gene accession number is NM_000020.

Assay Conditions: Assay was done in Kinase
buffer containing 1 mM DTT using Casein
substrate (0.2 mg/ml) and 20 µM ATP.
Reaction was done at 30°C for 45 min. The
amount of ATP transferred from
autophosphorylation as well as substrate
phosphorylation was calculated using KinaseGlo
reagent (Promega).
Formulation: 40 mM Tris-HCl pH 8.0,
110 mM NaCl, 2.2 mM KCl, 0.04% Tween20,
0.2mM EDTA, 3mM DTT and 20%
glycerol.

Storage / Stability: >6 months at -80°C. Application(s): Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
