Assay done using ELISA technology. 50 µl reaction mix containing HEPES-based buffer and 0-4 µg FTO is added to the wells coated with the m6A substrate. Incubate for 2 hr. at 37°C. Add antibody against N6-methyladenosine, incubate 1 hr. Then, add secondary HRP-labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminescence.

40 mM Tris-HCl pH 8.0, 110 mM NaCl, 2.2 mM KCl, 3 mM DTT and 20% glycerol.

Aqueous buffer solution

At least 6 months at –80°C.

Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
