Add 20-40 ng of BCL2L2 to 20 μl reaction mix containing 130 nM MCL1 substrate, anti-His Tb-labeled donor, and dyelabeled acceptor. Incubate 1h at room temperature. Read TR-FRET (λexc=340 nm, λem1=620 nm and λem2=665 nm).

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 0.04% Tween-20, 20% glycerol, 3mM DTT and Imidazole

At least 6 months at –80°C

Useful for the study of enzyme kinetics, screening inhibitors, selectivity profiling, and as a positive control for western blot studies.
