25 mM Tris/HCl, pH 8.0, 137 mM NaCl, 2.7 mM KCl, 1 mM MgCl₂, 0.1 mg/ml BSA, 200 µM SIRT5 substrate 5 (Catalog #50401), 500 µM NAD+ and Sirtuin 5 (15.6 - 1000 ng). Incubation condition: 30 min at 37°C followed by treatment with 50 µl of SIRT developer (Catalog # 50089) for 15 min at RT. Fluorescence intensity is measured at ex360/em460.

45 mM Tris-HCl, pH 8.0, 124 mM NaCl, 2.4 mM KCl, 18 mM glutathione and 10% glycerol.
