Assay was performed using BRD9 Inhibitor Screening Assay Kit, Cat. # 32519. BRD9 protein is incubated with biotinylated acetyl-histone peptide substrate BET Bromodomain Ligand in BRD9 assay buffer in a 10 µl reaction for 30 min at RT. AlphaLISA^® ® GSH acceptor beads (PerkinElmer) were then added, and the mixture was incubated for 30 minutes at room temperature (RT). Alphascreen^®® Streptavidin-conjugated donor beads (PerkinElmer) were then added. After 10 minute incubation at RT, Alphacounts are measured.

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, and 20% glycerol.

Aqueous buffer solution

>6 months at –80°C.

Useful for the study of bromodomain binding assays, screening inhibitors, and selectivity profiling.
