Background: POLG is the catalytic subunit of the mitochondrial DNA polymerase. The catalytic subunit contains three enzymatic activities, a DNA polymerase activity, a 3’-5’ exonuclease activity that proofreads misincorporated nucleotides, and a 5’-dRP lyase activity required for base excision repair. Mutations in the POLG gene are associated with several mitochondrial diseases, including Alpers' disease, ataxia-neuropathy disorders, and dominant and recessive forms of progressive external ophthalmoplegia.
Description: Human DNA polymerase γ (gamma), also known as Pol gamma and POLG (GenBank Accession No. NM_002693), a.a. 2-1239 (end), with N-terminal His-tag and N-terminal FLAG-tag, MW=141 kDa, expressed in a baculovirus infected Sf9 cell expression system.
Synonym(s): DNA polymerase gamma, Pol gamma, POLG
Formulation: 40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 80 µg/ml FLAG peptide, 0.04% Tween-20, and 20% glycerol. Format: Aqueous buffer solution
Storage / Stability: >6 months at -80°C. Application(s): Useful for DNA synthesis and DNA repair, as an antigen for Western blotting, and for inhibitor screening.
