Background: The Wnt Signaling Pathway TCF/LEF Reporter (Luc) – HEK293 Cell Line (Lithium-Independent)
is designed for monitoring the activity of the Wnt/b-catenin signaling pathway, without the
requirement for external LiCl. The Wnt pathway controls a large and diverse set of cell fate
decisions in embryonic development, adult organ maintenance and disease. Wnt proteins bind
to receptors on the cell surface, initiating a signaling cascade that leads to stabilization and
nuclear translocation of b-catenin. b-catenin then binds to TCF/LEF transcription factors in the
nucleus, leading to transcription and expression of Wnt-responsive genes.
Description: The Wnt Signaling Pathway TCF/LEF Reporter (Luc) – HEK293 Cell Line contains a firefly luciferase gene under the control of TCF/LEF responsive elements stably integrated into HEK293 cells. This cell line is activated by Wnt3a alone, without the need for Lithium Chloride to boost the signal. It is validated for response to the stimulation of mouse Wnt3a and to treatment with an inhibitor of the Wnt/b-catenin signaling pathway.
Materials Required But Not Provided: • Mouse Wnt3a (R&D Systems 1324-WN) Supplied As: Each vial contains ~1.5 X 10^6 cells in 1 ml of 10% DMSO.
Mycoplasma Testing: The cell line has been screened using the PCR-based VenorGeM Mycoplasma Detection kit (Sigma Aldrich) to confirm the absence of Mycoplasma species.
Growth Media: Thaw Medium 1 (BPS Cat. #60187): MEM medium (Hyclone #SH30024.01) supplemented with 10% FBS (Invitrogen #26140-079), 1% non-essential amino acids (Hyclone #SH30238.01), 1 mM Na pyruvate (Hyclone #SH30239.01), 1% Penicillin/Streptomycin (Hyclone SV30010.01). Format: Aqueous solution containing DMSO.
